Dissertation is devoted to the identification and characterization of novel human genes using the nucleotide sequences of NotI-linking clones, which are associated with CpG-islands. Three novel human genes have been identified: potassium channel Kv1.7 gene, hUNC93 gene and kinase MLK4 gene. The isolation has been performed with the following methods: PCR, cDNA library screening, RT-PCR and RACE. It has been determined the optimal concentration of formamide and glycerol in the reaction mixture for GC-rich template sequencing. The level of expression in different tissues, genomic structure and chromosomal localization have been determined for each identified gene. It has been identified domains and motifs in the deduced amino acid sequences, corresponded to the isolated genes. Using the data concerning the function of corresponded homologues and orthologs, the role of gene products in the human organism has been predicted.
